Common use

Tizacare is commonly prescribed for the management of osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis in adults. It is also indicated for the treatment of acute pain, such as that associated with menstrual cramps, and for the reduction of adenomatous colorectal polyps in specific high-risk patient populations, under specialist care. Its use is generally reserved for cases where non-pharmacological interventions and first-line treatments have provided insufficient relief or are not tolerated.

Dosage and direction

The recommended dosage of Tizacare must be individualized based on the condition being treated and patient response. For osteoarthritis, the usual dose is 200 mg per day, administered as a single dose or divided into two 100 mg doses. For rheumatoid arthritis, the typical dose is 100 mg to 200 mg twice daily. For acute pain or primary dysmenorrhea, an initial dose of 400 mg may be used, followed by an additional 200 mg dose if needed on the first day, with subsequent doses of 200 mg twice daily as required. It should be taken with a full glass of water, and may be taken with or without food to minimize potential stomach upset. Do not crush, chew, or break the capsules.

Precautions

Patients should be advised to use the lowest effective dose for the shortest duration consistent with individual treatment goals. Regular monitoring of blood pressure, renal function, and for signs of gastrointestinal bleeding is recommended, especially in elderly patients or those with a history of peptic ulcer disease. Use with caution in patients with fluid retention, hypertension, or heart failure. Patients should be informed about the signs and symptoms of cardiovascular events, gastrointestinal events, and serious skin reactions, and instructed to seek immediate medical attention if they occur.

Contraindications

Tizacare is contraindicated in patients with known hypersensitivity to celecoxib or any component of the formulation. It should not be used in patients who have experienced asthma, urticaria, or other allergic-type reactions after taking aspirin or other NSAIDs. Contraindicated in the setting of coronary artery bypass graft (CABG) surgery. Should not be used in patients with severe hepatic impairment or in those with inflammatory bowel disease. Not recommended for use during the third trimester of pregnancy.

Possible side effect

Common side effects may include headache, dizziness, insomnia, abdominal pain, diarrhea, dyspepsia, flatulence, nausea, and peripheral edema. Less frequently, rash, elevated blood pressure, and tinnitus may occur. Serious but rare side effects can include myocardial infarction, stroke, gastrointestinal bleeding, ulceration, or perforation, hepatotoxicity, severe skin reactions such as Stevens-Johnson syndrome, acute renal failure, and anaphylactic reactions. Patients should report any unusual or severe symptoms to their healthcare provider promptly.

Drug interaction

Tizacare may interact with ACE inhibitors, angiotensin II receptor blockers, diuretics, and other antihypertensive agents, potentially reducing their effectiveness. Concurrent use with aspirin or other NSAIDs may increase the risk of gastrointestinal adverse events. It may enhance the effects of warfarin and other anticoagulants, increasing the risk of bleeding. Interactions with fluconazole may increase celecoxib plasma concentrations. Lithium levels may be increased when taken concomitantly. Always inform your healthcare provider of all medications, including over-the-counter drugs and supplements.

Missed dose

If a dose is missed, it should be taken as soon as remembered, unless it is almost time for the next scheduled dose. In that case, skip the missed dose and resume the usual dosing schedule. Do not double the dose to catch up. Maintaining a consistent dosing schedule is important for therapeutic effectiveness.

Overdose

In case of suspected overdose, seek immediate medical attention or contact a poison control center. Symptoms of overdose may include lethargy, drowsiness, nausea, vomiting, epigastric pain, gastrointestinal bleeding, hypertension, acute renal failure, and respiratory depression. Supportive and symptomatic treatment is recommended; there is no specific antidote. Hemodialysis is not effective in removing celecoxib due to high protein binding.

Storage

Store at room temperature between 20°C to 25°C (68°F to 77°F), with excursions permitted between 15°C to 30°C (59°F to 86°F). Keep in the original container, tightly closed, and protect from light and moisture. Do not store in the bathroom or near sinks. Keep out of reach of children and pets. Do not use beyond the expiration date printed on the packaging.
